Role summary: Incumbent will provide direction, oversight and support for generation of antibodies as tool reagents for evaluating bacterial vaccines including antibody characterization and their use in development of preclinical assays. The incumbent will be responsible for the generation and development of different types of antibodies with the clear focus of monoclonal antibodies originated from B-cells with respective functional activity; primarily performing bench work. Duties will include screening, expression, purification and their characterization with various bioanalytical methodologies and functional assays. Supporting the generation of antibodies based on the hybridoma technology will be a part of the role. An important role is guidance of more junior people in the lab. The ability to work in teams, write reports, standard operation procedures (SOPs) and publications for peer review journals is required. The incumbent will be involved in project management, being closely aligned with bacterial vaccines, vaccines research and development management and multiple program teams. Role responsibilities: Isolation of human monoclonal antibodies from peripheral blood B cells including cloning, expression and characterization of these recombinant antibodies showing neutralizing activity performing the entire bench work by herself/himself. Furthermore generation of monoclonal antibodies based on the hybridoma technology for multiple projects in the anti-infective and bacterial field, including screening and characterization with ELISA, Octet/Biacore, FACS, immune blot and various biophysical methodologies. Purification of the monoclonal antibodies using state-of-the-art chromatography methods. Data interpretation and documentation including handling of the entire antibody inventory. Communication of data to involved program leads and team members. Research and design experiments to resolve technical problems Performing/conducting the experiments by herself/himself in the designed laboratories. Transfer of developed processes to team members combined with their training/education. Improving current processes in the direction of automation to increase throughput. Understanding of relevant work from the scientific community and using the information to improve internal processes and/or establish new procedures/technologies. Design and carry out assays for antibody characterization. Perform other duties as assigned. Qualifications: Advanced scientific degree (PhD, MD, or equivalent) + 4 years experience, preferably in a pharmaceutical or biotechnology environment, MS +9 years experience in a field of science relevant to the position for external/internal candidates. Laboratory experience in a relevant field of science. The incumbent will be trained on required Policies, Practices, and Procedures needed to support their work responsibilities. Proficiency in the use of Word processing, Excel, PowerPoint and relevant scientific software is desirable. Good verbal and written communication skills are required. Hands-on skills in protein expression, purification and characterization using biochemical/biophysical and functional assays such as SDS-PAGE, immune blot, size-exclusion-HPLC, ELISA and neutralization assays. Interest to learn new techniques. Working experience with mammalian cells and the ability to write protocols independently. The incumbent is responsible for planning her (his) daily workflow to complete the tasks/experiments outlined by the manager. Physical/mental requirements: Physical positions: work in the lab is partly done sitting but mostly standing whereas sitting is required for office work (computer and writing of laboratory notebooks). Additionally, sitting for sterile cell culture manipulations using Biological Safety Cabinet is mandatory.
