The Gene Therapy Translational Research group is part of Pfizer's Rare Disease Research Unit (RDRU) focused on gene therapy specific platform technologies towards the delivery of IND-approved vectors for testing in clinical trials. This Senior Scientist will be part of an interdisciplinary team partnering closely with the RDRU Biology and Gene Therapy groups as well as colleagues in other Pfizer disciplines contributing to translational preclinical packages related to novel gene therapies. This bench Scientist will be part of an interdisciplinary team and contribute to preclinical data required to advance Pfizer's gene therapy-based portfolio. This Senior Scientist will develop novel 3D, organoid and explant cell systems in support of AAV cell based assays. The Sr. Sci. will have advanced expertize in at least of the areas (3D cultures, cultures with feeder cells and matrix proteins, organoid cultures, primary stem cells, explants) and working knowledge of the remaining areas. Sr. Sci. will work independently, use scientific judgment in making decisions within the area of expertize, proactively gather inputs from colleagues, develop novel processes, and is expected to provide significant scientific expertise to the team. MAIN RESPONSIBILITIES Establish novel cell systems including 3D cultures, organoid cultures, and explant cultures. Validate the utility of the novel cell culture systems for in vitro characterization of AAV vectors. Utilize advanced cell culture methods to develop in vitro models of human disease using cells extracted from patients. Perform cell based assays related to delivering AAV gene therapy vectors for testing in clinical trials in therapeutic areas including liver, muscle and CNS targeting. Partner with colleagues working on Gene Therapy projects, e.g. RDRU Biology, I&I RU and IMRU colleagues, PDM, PharmSci, CDX and CAG colleagues. Deliver high-value data and lead in vivo aspects of AAV-based gene therapy projects in the RDRU pipeline. Evaluate and engage external collaborators and Contract Research Laboratories (CROs). BASIC QUALIFICATIONS Ph.D. in life sciences with 0-2 years of postdoctoral experience or Master's Degree with 2-4 years of academic or industry research experience Scientific training and background in the fields of cell biology or molecular biology. Cell culture expertise with 3D cultures, organoid cultures, or explanted tissues is required. Hands on expertise with isolation, cultivation, and differentiation of the primary cells derived from blood, liver, muscle, or brain (spinal cord) is required. Experience with additional tissues is a plus. Evidence of scientific accomplishments in the form of publications and presentations. Exceptional communication skills and demonstrated ability to work effectively as part of a team.
